Identification and Outlining
DIAS has the ability to open, save and analyze QuickTime® movies, Tiff stacks, as well as movies in its own format. Once the user has captured the movies or images of interest to their computer, DIAS can be used to automatically outline the cell perimeters. DIAS provides two unique automatic and manual methods for outlining. With all four methods of outlining, the user retains complete control over the process.
Determining Paths and Shapes
Once DIAS has outlined all of the objects of interest in a movie or image, it creates a file that is an abstraction of the original movie. Subtracting all backround information, leaving only the outlines and paths of the objects of interest, DIAS saves this information in a path file. DIAS computes a number of parameters based upon the area and contour of expansion and contraction zones over time. The user may window any portion of the cell, e.g., an expanding pseudopod or lamellipod, and quantitate the dynamics of extension and retraction over time.
Computing Parameters
DIAS automatically calculates more than 30 predefined parameters of motility and dynamic morphology for a single cell, or a population of cells. These calculations are based upon the dynamics of the cell center, and the changes in cell contour over time. In addition, the user may create additional parameters, using an advanced RPN calculator built into the software.
Displaying Data
DIAS can display data as graphs or as data sets, these graphs can be exported as images or as data. DIAS also exports all data into a Microsoft Excel compatible format. Anything that is displayed on the screen can be exported to either an image file, or a Quicktime movie.
Image Processing
DIAS also has advanced image processing capabilities. These transformations can be done to single images, or entire movies. Sharpen, blur, contrast/brightness adjustment, image rotatation and many other filters can be applied, directly within DIAS.
